Gene therapy: Editing out genetic damage

Jan. 31, 2013 ? New design guidelines from researchers in Singapore simplify the development of targeted therapies for muscular dystrophy and other diseases.

The dystrophin protein offers critical support to muscle fibers. Mutations affecting dystrophin's expression cause the muscle-wasting disease muscular dystrophy. In Duchenne muscular dystrophy (DMD), these mutations take the form of small sequence changes that make much of the dystrophin gene (DMD) untranslatable, yielding nonfunctional protein or no protein at all.

Therapies based on a strategy known as 'exon skipping' could undo the damage from these mutations. Development of such treatments is set to accelerate, thanks to research by a team led by Keng Boon Wee of the A*STAR Institute of High Performance Computing and Zacharias Pramono of the National Skin Centre in Singapore.

Proteins are translated from messenger RNA transcripts of genes; however, only certain RNA regions -- known as exons -- actually encode protein, and these are enzymatically spliced together prior to translation. Several clinical studies have demonstrated that small 'antisense oligonucleotide' (AON) molecules that bind mutated DMD exons can induce elimination of those defective exons during splicing, yielding shorter but largely functional versions of dystrophin. "We are cautiously optimistic that AON-induced exon skipping could be the first effective therapy for DMD patients," says Wee.

Unfortunately, DMD arises from many different mutations, and targeted AON design remains a time-consuming, trial-and-error process. To address this challenge, Wee and Pramono sought to define the characteristics of AONs that efficiently promote exon-skipping. They used computational analysis to zoom in on exonic sequences that coordinate splicing. They also identified regions of suitable length within dystrophin RNA transcripts that span these sequences and would be accessible to AONs in living cells.

The researchers thus derived a set of guidelines enabling them to effectively design AONs that targeted nine different exons affected in DMD patients. For each exon, at least one AON proved capable of boosting dystrophin expression to clinically relevant thresholds in cultured muscle cells (see image). "Our proposed set of factors resulted in a reasonable success rate of designing efficient AONs -- 61% versus 38% using semi-empirical methods," says Wee. Clinical studies have already demonstrated the promise of efficient exon skipping in treating DMD patients.

Wee notes that other diseases arising from abnormal RNA processing could also benefit from this approach. However, his team is also exploring this method as a general strategy to abort production of disease-causing proteins in cancer and other conditions. "In contrast to small-molecule inhibitor drugs that can target only about 10% of the human genome, this approach could downregulate most human genes," Wee says.

Lil Wayne has yet to watch Nicki Minaj on 'Idol'

METARIE, La. (AP) ? "American Idol" picked Nicki Minaj as a judge to draw in viewers, but she's actually discouraged at least one person from watching it ? her mentor, Lil Wayne.

The rap star admitted Friday that he has yet to see "Idol" this season ? or any other season.

"Actually, I've never watched it," he said, a bit sheepishly.

Part of the reason he hasn't tuned in to see Minaj is because she didn't exactly sell the show to Lil Wayne.

"When I talk to her she's always tells me, 'You don't wanna watch that, you're not going to be into it,'" he said.

"She knows me. ... It's kind of like a catfight where they be going back and forth type of thing, I'm not into all that," he said of the judging panel, which includes Mariah Carey, with whom Minaj had a much-publicized argument.

"The talent on that is pretty good I heard, though," he said.

Lil Wayne spoke Friday after signing autographs for more than 100 fans who were at a suburban New Orleans Macy's to support his Trukfit clothing line; a couple hundred more waited outside for a glimpse of the star.

Wayne said he's hoping to soon make Trukfit for women and children. Now, it's just a men's line. Part of the problem is finding the right designer.

"They send everything, every design to me, and I approve or decline, and the designer is extremely OK (with that). ... That person may have designed something, and I may say I hate it. I get no flack about it, and women are different," he said, laughing. "So we're trying to find the perfect female designer who has no problem with me declining what I don't like."

Lil Wayne, of course, still has his day job: His new album, "I Am Not A Human Being II," is due out March 26. When asked what fans could expect, he said, laughing: "Never have expectations for me, like never, because I probably will not meet your expectations. ... Just enjoy what you get, hopefully."

Hotel prices rising in Latin American cities | Buying Business Travel

Latin American cities have seen the biggest rises in hotel prices over the last year, according to figures from HRG.

Cities such as Rio de Janeiro and Sao Paulo recorded rate rises of 19 and 16 per cent respectively in local currency, although UK corporate buyers were largely protected from these hikes due to exchange rates.

Moscow continues to be the most expensive city for hotels with an average room rate of ?261.38 in 2012, although this was only up by 1 per cent on the previous year. The Russian capital has had the highest accommodation prices for nine consecutive years.

The next most expensive cities for hotels were Lagos, New York, Hong Kong and Zurich. London only ranked in 17th place in the list of the top 55 cities around the world with an average rate of ?164.30 up by 5 per cent on 2011.

HRG?s group commercial director Stewart Harvey said: ?HRG?s Hotel Survey shows that regional trends are becoming less relevant as individual cities become bigger players in an ever-shrinking world. ?

?Though the overall pattern seems to be one of rising average room rates, it?s interesting to see just how varied the picture is within regions, and even within different countries.?

Outside of London, there were also rate increases in Aberdeen, Liverpool and Belfast. But prices in Manchester and Edinburgh went down slightly.

Most US cities saw higher hotels rates with San Francisco and Atlanta recording rises of 11 and 9 per cent respectively on the back of strong convention business. But the capital Washington DC saw rates fall by 13 per cent.

The picture was ?patchy? across Europe - rates were flat in major destinations such as Paris and Amsterdam as measured in local currency, while German cities such as Frankfurt, Munich and Dusseldorf saw small rises.

The report also found that most major Indian cities had seen a decline in hotel rates due to the economic slowdown in the country.

Harvey added: ?For several years now, we?ve seen corporates becoming increasingly cost aware and have implemented greater control over travel options. This focus on prudency is expected to continue.?

New protocol recommendations for measuring soil organic carbon sequestration

Feb. 1, 2013 ? Increased levels of greenhouse gases, particularly carbon dioxide (CO2), have been associated with the burning of fossil fuels, deforestation, cultivation of grasslands, drainage of the land, and land use changes. Concerns about long-term shifts in climate patterns have led scientists to measure soil organic carbon (SOC) in agricultural landscapes and to develop methods to evaluate how changes in tillage practices affect atmospheric carbon sequestration. University of Illinois professor of soil science Kenneth Olson has used data collected over a 20-year period at Dixon Springs, Ill., to develop a new protocol for more accurately measuring the carbon removed from the atmosphere and subsequently sequestered in the soil as SOC.

"Many experiments comparing no-till to conventional tillage on similar soils have shown no-till to have higher levels of soil organic carbon," Olson said. "So we know in general that no-till is often better than conventional tillage at building or retaining more of the organic matter in the soil, which is important to crop productivity. However, this does not mean that no-till is necessarily sequestering atmospheric carbon. It is often just losing carbon at a lower rate than conventional tillage." This unexpected discovery was the result of Olson's use of a pre-treatment SOC measurement method that compares change in soil organic carbon over time on the same plots using the same tillage methods. "This protocol does not assume that soil carbon pools are at steady state (remain the same over time), but measures SOC at the beginning of an experiment, at intervals during, and at the end of the experiment," Olson said.

"Comparison studies with one treatment as the baseline (usually conventional tillage) or control and other tillage such as no-till as the experimental treatment should not be used to determine SOC sequestration if soil samples are only collected and tested once during or at the end of the study," Olson said. The comparison method assumes the conventional tillage baseline to be at a steady state and having the same amount of SOC at the beginning and at the end of the long-term study, and this may not be true. No-till as the experiment treatment needs to be compared to itself on the same soils over time to determine if SOC sequestration has really occurred.

Olson compared two decades of data from previously eroded Grantsburg soils on 6 percent slopes to a 30-inch depth with low SOC content in an attempt to quantify the amount and rates of SOC sequestration, storage, retention, or loss as a result of a conversion from conventional tillage to a no-till system. Olson used both the comparison and the pre-treatment SOC measurement methods on the same plot area. His analysis revealed conventional tillage and no-till plot areas had less carbon (C) at the end of the study than at the beginning using the pre-treatment SOC method. According to the comparison method, no-till sequestered 4.1 tons of C per acre for a 17 percent gain during the 20 years of the study. However, the pre-treatment SOC method showed that the no-till plots actually lost 3.1 tons of C per acre, a 13 percent loss in 20 years. Thus, no SOC sequestration had actually occurred during the Dixon Springs study.

There were three major reasons why the comparison study approach was the wrong method for measuring C sequestration on the Dixon Springs plot area. First, the conventional tillage plots were not at steady state and actually lost 30 percent of the C in 20 years due to erosion and SOC-rich sediment being transported off the plots. Second, when the no-till and conventional tillage plots were sampled only once, it was not possible to determine the rate of change over time. Last, the effect of tillage equipment breaking down the soil aggregates increased the carbon available to microbial decomposition and the release of C to the atmosphere as CO?.

"Field experiments must be designed to more carefully measure, monitor, and assess internal and external inputs," Olson said. "The amount of SOC loss from soil storage during the time of the experiment needs to be subtracted from SOC gains to determine the change in net SOC storage. Further, soil laboratory and field methods for quantifying SOC concentration must be refined to reduce under- and over-estimation bias."

Olson also recommends that the definition of SOC sequestration include a reference to the land unit. "Soil organic carbon sequestration is currently defined as the process of transferring CO? from the atmosphere into the soil through plants, plant residues, and other organic solids that are stored or retained as part of the soil organic matter (humus). The retention time of sequestered carbon in the soil (terrestrial pool) can range from short-term (not immediately released back to the atmosphere) to long-term (millennia) storage," Olson said. The SOC sequestration process should increase net SOC storage during and at the end of a study to above the previous pre-treatment baseline levels and result in a net reduction in the atmospheric CO? levels. I believe that the phrase 'of a land unit' needs to be added to the definition to add clarity and to exclude the loading or adding of organic C derived naturally or artificially from external sources," Olson suggested.

Olson concluded by saying that carbon not directly from the atmosphere and from outside the land unit should not be counted as sequestered SOC. The definition of SOC sequestration as defined with borders would mean any C already in storage and transported or redistributed to the plot area or field would have to be accounted for and does not qualify as sequestered SOC.

"Any manure from outside the plot area or SOC-rich sediments transported and deposited from adjacent upland are just redistributed or transported C and not really sequestered SOC," Olson said. "That C was already in storage and may in fact be released back to the atmosphere if applied to the plot. For example, decomposing manure loaded on a land unit increases the return of CO?to the atmosphere and does not result in a depletion of atmospheric CO?, which is the real goal. Because we often lack the ability to directly measure the total change in the atmospheric CO? as a result of C loading on a plot or field, we indirectly estimate it by measuring the change in amount of SOC being stored in the land unit.

"These proposed protocols are necessary to move the science forward and to attempt to address future predicted climate trends," Olson said. "The amount of SOC sequestered as a result of alternative agricultural systems such as no-till and its effects on net SOC storage changes in the soil over time and the SOC released to the water and atmospheric pools need to be measured or calculated."

Olson said that any future Cap and Trade program will require SOC sequestration protocols to be established. The method of measurement is critical if SOC sequestration is to be verified. "If landowners are to truly sequester SOC, they must be able to prove that net carbon gains have occurred over time in their fields and that the increased SOC remains permanently stored in their soil," Olson said.

I also grow my own food
Ive been feeding shrimp pellets lately as my shrimp are not breeding quite fast enough but I have a colony of shrimp I use for feed
I currently feed a combo or frozen blood worms, shrimp pellets, and live shrimp

If you want to continue to use feeder fish I would suggest setting up a quarantine tank, keep them there for at least a few weeks before feeding to the lotls
Any fish from any store needs to be placed in quarantine before being introduced to your pets, any fish from any store has the potential to carry any number of diseases or fungus

as for your question I cant say for sure how much food would satisfy your lotl, I feed mine 2 or 3 times a week but if I go a extra day or two without feeding they start nipping at each other
mine arent quite a year old yet so I suspect they are more hungry than older lotls

Don?t you just hate it when you join a game online and the first thing you see in chat is a language you don?t understand? Yeah, me too. Every Dota 2 player out there see?s this in almost every public match and believe it or not, we still don?t know what the hell they are saying. There?s a growing issue when it comes to playing online and the language barrier on European servers, I think something should be done about that.

Whether you?re playing an MMO or MOBA, you have probably experienced the frustration of people in your party (group) being blatantly rude and having a whole conversation in French, German, Polish or Russian (yes, there are probably some other languages in the mix too). And even if you, as an English speaking South African, American or any other nationality speaking the universal language, ask for these gamers to speak English, they just flat-out ignore you and carry on.

This creates not only a bad atmosphere but obviously a lack of communication and more often than not, causes the group to fail at whatever they are doing. Try getting some boss tactics in order while in an instance (dungeon) or communicating in a Dota 2 game while 3/5 of the party members refuse to speak English, it just doesn?t work. The problem isn?t that people speak different languages, but that this defeats the public match making feature in games when it results in failure and arguments or people just quitting games. It?s not enjoyable and it?s now a part of our everyday lives. The solution is that we need a way to communicate with each other without conflict and only having one option: to only speak English.

My suggestion to developers is to include an in-game translator to the chat boxes. ?Some might argue that this is a violation of privacy, because maybe the people speaking the other language don?t want you to see what they are saying. My response to that is: if you are typing anything into a public party chat, you don?t give a crap since any one can use Google translate to know what you?ve just typed, and you also don?t know enough about the people in the party to know that they don?t understand the language you are typing in. ?It would be easy for us to say ?if you want to type in your language, use personal messaging?, but we all know that?s not going to happen.?It?s a lot easier to say ?if you don?t want me to know what you are saying?, don?t type it in public.

Having an in-game translator in my mind cannot be impossible to code into a game. If I can connect to Facebook and Twitter in Rift (the MMO) and have the game automatically tweet and post achievements or upload screenshots, then I?m sure it can be coded to grab text, detect the language and translate it. This means that if I?m for example French and I don?t understand English, I can translate the English text in-game and no one has to sit through the frustrating experience of not understanding each other.

Bottom line, if you?re giving us the option of public match making, give us the option to understand each other and break that language barrier that makes the whole experience more painful than ripping off that piece of skin right next to your finger nail.
